What is LNA?
LNA is a novel type of nucleic acid analog that contains a
2'-O, 4'-C methylene bridge. This bridge restricts the flexibility of the ribofuranose ring and locks the structure into a rigid bicyclic formation, conferring enhanced hybridization performance and exceptional biostability

LNA oligos are innovative oligonucleotides that contain Locked Nucleic Acid (LNA) to provide superior hybridization characteristics and enhanced biostability compared to conventional DNA oligonucleotides. LNA oligos can be designed to enhance a wide variety of genomic applications and technologies that rely upon the use of oligonucleotides.
- Deprotected and desalted
- Purified by PAGE, RPC or RP-HPLC upon request
- Delivered in solution in de-ionized water
- Shipped within 3 to 5 working days of receiving your order, pending successful QC validation
- Systematic QC by PAGE or mass spectrometry analysis and - depending upon the nature of the oligonucleotide requested - by RP-HPLC or IE-HPLC
LNA oligos are composed of either 100% Locked Nucleic Acid (LNA) bases or a mixture of LNA and DNA bases. However, only oligonucleotides that are between 7 and 15 mers in length can be made up of 100% LNA. If an oligonucleotide is longer, the 100% LNA oligo may bind to itself (self complement) rather than to the target sequence. Therefore, for oligonucleotides longer than 15 mers, we recommend decreasing the LNA content as the oligonucleotide increases in length, as follows:

Guaranteed Yields, Labels, Modifications and Turn-Around Time for LNA oligos

LNA oligonucleotides can be designed to enhance a wide variety of genomic applications and technologies that rely upon the use of oligonucleotides:
- Hybridization probes for the detection of nucleic acids and genotyping e.g. modified Molecular beacons
- Padlock probes, modified Taqman™ probes and probes for in situ hybridization
- Capture probes for target enrichment, sample preparation and affinity chromatography
- Allele-specific PCR1
- Target validation through antisense technology
- Oligonucleotides for therapeutic diagnostic applications, such as antisense oligonucleotides, aptamers, ribozymes, decoy oligonucleotides and siRNA
- LNA oligonucleotides have been applied in strand invasion techniques
In most respects, LNA oligonucleotides can be used in the same manner as DNA oligonucleotides: it is soluble in aqueous buffers, can be ethanol precipitated, dried and resuspended, and analyzed by gel electrophoresis, HPLC and MALDI-TOF.
Specifications of LNA Oligos
| Quality control | All our oligonulceotides undergo vigorous process monitoring and strict quality control. Length and labeling are systematically controlled by PAGE or mass spectrometry analysis and - depending on the nature of the oligonucleotide requested, by RP-HPLC or IE-HPLC. Quantity is systematically validated by UV absorbance at 260 nm. |

| Storage and stability | Although oligonucleotides are stable in solution at 4¡É for up to 2 weeks, Proligo recommends storage should be at -20¡É. Repetitive freeze-thaw cycles should be avoided by storing as aliquots. Storing at concentrations above 20¡É is recommended. Oligonucleotides with fluorescent labels should be protected from light. Proligo guarantees its oligonucleotides for six months, stored under the above conditions. |
